To produce Penicillin using Penicillium chrysogenum.

Aim : To produce Penicillin using Penicillium chrysogenum.

Requirements :

48 hr culture of Penicillium chrysogenum
Inoculum and Production ( complex) medium
Lactose 20.0g
Corn step liquor 20.0g
Pottasium dihydrogen
Phosphate 0.5g
Mag. Sulphate 0.25g
Sodium nitrate 3.0g
Zinc Sulphate 0.04g
Calcium Carbonate 2.75g
Distilled water up to 1.0 lit
PH 4.7

Procedure:
Spores from a 48 hr slant culture of P. chrysogenum were transferred to 50 ml of sterile inoculum medium of the above composition. The flask was incubated on a shaker for 48 hr. and 10 % level of the inoculum was transferred to 200ml of production medium taken in a one-liter flask. Termination was carried out on a rotary shaker for one week. Samples were drawn at 48, 60, 72, 84, 96, 120, 144 and 168 hr. and PH and PCV were determined for each. Potency of each sample was tested microbiologically using the standard graph.

Result:
The given strain produced a maximum yield of 224.8 unit /ml at 84 hr.
